Nuclear and Cytoplasmic Degradation
We study the proteolytic events in the cytosol and the role they play supplying precursors to the MHC class I antigen processing pathway. Our aims are: 1) Identification of substrates of the ubiquitin/proteasome pathway, and determination of whether newly synthesized proteins constitute a significant fraction of these substrates. 2) Determination of the relationship between these substrates and peptides presented by MHC class I. 3) Identification of ubiquitin/proteasome-independent proteolytic pathways. Our data seem to discard the suggested role for the enzyme tripeptidyl peptidase II. Our data point instead to a prominent role of macroautophagy.
